Coenzyme Q10 exists in 3 redox (reduced/oxidized) states:  fully oxidized (ubiquinone), semiquinone (ubisemiquinone), and fully reduced (ubiquinol).  All 3 forms are essentially bio-equivalent, though  ubiquinol has 10% greater absorption.   Coenzyme Q10 is abbreviated as CoQ10, CoQ, or Q10 is a 1,4-benzoquinone, where “Q” refers to the quinone chemical group, and 10 refers to the number of isoprenyl chemical subunits in its tail.  It was discovered in 1954, and in the 1980s there was a steep rise in clinical trials.  This oil-soluble, vitamin-like substance is present in most eukaryotic cells, primarily in the mitochondria, which is found in every cell in the body [also in LDL, HDL, & the cell membrane at].  It is a component of the electron transport chain and participates in aerobic cellular respiration, generating energy in the form of ATP. Ninety-five percent of the human body’s energy is generated this way.  Those organs with the highest energy requirements—such as the heart, liver, [brain], and kidney—have the highest CoQ10 concentrations.  The capacity of this molecule to exist in a completely oxidized form and a completely reduced form enables it to perform its functions in the Electron Transport Chain (ETC) and as an antioxidant respectively.  CoQ10 is fat-soluble and is therefore mobile in cellular membranes; it plays a unique role in the ETC.  In the inner mitochondrial membrane, electrons from NADH and succinate pass through the ETC to the oxygen, which is then reduced to water. The transfer of electrons through ETC results in the pumping of H+ across the membrane creating a proton gradient across the membrane, which is used by ATP synthase (located on the membrane) to generate ATP[1].  This is crucial in the process, since no other molecule can perform this function. As an antioxidant, Q10’s also improves health.  The antioxidant nature of Q10 derives from its energy carrier function.  As an energy carrier, the CoQ10 molecule is continually going through an oxidation-reduction cycle. As it accepts electrons, it becomes reduced. As it gives up electrons, it becomes oxidized. In its reduced form, the CoQ10 molecule holds electrons rather loosely, so this CoQ10 molecule will quite easily give up one or both electrons and, thus, act as an antioxidant”(Wiki).  It is considered the best of anti-oxidants because it is distributed widely in the body and naturally serves this function with a low energy barrier, and mechanism for recycling.  Long-term usage is required for benefits, especially for reduction in damage to proteins which cause age related chronic conditions, and LDL that starts atherogenesis that causes heart disease--summary.  Peak blood level is in 2-6 hours, and second peak at about 24 hours; half-life is 33 hours. The synthesis of the precursor of coenzyme Q10, mevalonate, is inhibited by some beta blockers (blood pressure-lowering medication), bisphosphonates statins, tricyclic antidepressants and other drugs. Q10 does not interact with drugs or have side effects, and has been shown safe at 2,400 mg & 3,600 mg. It is an important antioxidant, to which consistent with pharma, they oppose CoQ10 through denying its importance—profits first.

Mean reduction in migraine frequency after 1 month of treatment was 13.1% and this increased to 55.3% by the end of 3 months.

Statin induced Myopathy (muscle weakness and pain) and other side effects:  Statins inhibit the mevalonate pathway, which effects both the production of cholesterol and of Q10 by over 40%.  Thus statins reduce ATP, the body’s source for energy including those that drive bodily synthesis.  In the elderly (over 60) this effect is pronounced.   Reduction in Q10 (thus ATP) accounts for the long list and high frequency of side effects.  Statins are incompatible with the maximal exertion of elite athletes  Myopathy is ameliorated by Q10 supplementation.  Statins is poorly tolerated by the elderly.


Physical endurance:  Results indicate that acute and chronic supplementation of Q10 may affect acute and/or chronic responses to various types of exercise”; confirmed in a study of elite athletes, as “incompatible”.

Unfortunate there are no long-term clinical trials are population studies.  Negative studies are funded by pharma for financial gains.  However, positive claims are supported given its role as a ubiquitous antioxidant--see benefits above. For this reason it should be taken long-term starting by the age of 20, even though the decline in ATP due to damage won’t be noticed until the 6th decade, when endurance dramatically declines.   Q10 should be part of healthy diet by reducing sugars & carbs, exercise, learn of the corruption worked by Pharma, take aspirin, and post-menopause natural HRT.  Q10s usage is far greater in Japan and Europe.  JK takes300 mg daily and wishes he had started decades earlier.

Q10 (CoQ10):   recognized as the most effective antioxidant.  A number of major health conditions are caused by oxidative damage including hardening of the arteries for which Q10 is protective.   Q10 is found in every cell in the body because it is used by the mitochondria in the production of ATP from glucose.  ATP accounts for 95% of the body’s energy.   The antioxidant nature of Q10 derives from its energy carrier function in the production of ATP.   As an energy carrier, the Q10 molecule is continually going through an oxidation-reduction cycle.  It is the best of anti-oxidants because it is distributed throughout the body.  It is found in LDL (bad cholesterol) and thus protects by preventing oxidative damage to LDL which is the initial step in the process that results in hardening of the arteries (cardiovascular disease, CVD).  High blood pressure is a result of clogged, stiff arteries.  CVD causes over 80& of heart attacks and strokes.  It protects other tissues from oxidative damage and thereby reduces the risk for a number of serious conditions including Alzheimer’s & Parkinson’s diseases, CVD, and macular degeneration.  The second important function is protection of the mitochondria from damage from the reactive chemicals produced in the metabolism of glucose.    Long term usage of Q10 through protection of the mitochondria a greater production of ATP, thus greater endurance and improved health for the elderly.  It is particular beneficial for those with heart disease, hypertensions, diabetics; and for those taking statins (for cholesterol), bisphosphonates (for bones), or beta blocker (for hypertension), for these drugs partially block the bodily production of Q10 (40% for statins) and thus other products of the mevalonate pathway.  All these benefits from long-term Q10 add up to an increase in lifespan and greater endurance.  Q10 is not toxic:  a study found that daily dose of 3600 mg was tolerated by both the healthy and unhealthy.  Recommendation:  100 mg for children, being gradually increased to 300 mg by the age of 40--and its yearly cost through Costco is under $70 for Kirkland brand Q10.

As for a rebound effect from discontinuation of Q10, that hasn’t been reported, nor has the research addressed that possibility.  Since Q10 is at the end of a pathway which produces farnesyl-PP, and this is used to synthesize 7 essential families of compounds including heme A, Q10, sterol, cholesterol, and dolichols, the production of farnesyl-PP will not be significantly diminished by Q10 suppliment since it is only one of many end products.  Q10 production may be less for a few days if the supplement is stopped.  Farnesyl-PP is converted to Q10 where needed, thus serum level is naturally low and an increased level from exogenous source will have little effect upon the production of farnesyl-PP. 






Parkinson Disease: The plasma level reached a plateau at the 2400 mg/day dosage and did not increase further at the 3000 mg/day dosage.
